About RPM vs RCF in Centrifugation

RPM (revolutions per minute) describes how fast the rotor spins but does not tell you the actual force applied to the sample โ€” this depends entirely on the rotor radius. Two centrifuges running at the same RPM with different rotors will apply completely different forces to the sample.

RCF (Relative Centrifugal Force), expressed as ร—g, is the universally comparable unit because it is independent of rotor size. When following published protocols, always use RCF (ร—g) values to ensure reproducibility across different instruments.

Published protocols specify RCF (ร—g) not RPM, because RPM is meaningless without knowing the rotor. Convert to RPM only for your specific rotor.
๐Ÿ“ What is rotor radius?
The rotor radius (r) is measured from the centre of the rotor shaft to the tip of the sample tube. Check your centrifuge manual or rotor datasheet.
๐Ÿ”„ Average vs max radius
Some protocols specify average radius (midpoint of tube), others use maximum radius (tip of tube). Max radius gives maximum RCF; average radius gives average RCF.
๐ŸŒก๏ธ Temperature matters
Always specify temperature (4ยฐC for most biological samples). Heat generated during centrifugation can degrade sensitive proteins, RNA and cell viability.
