How to Use the Protein Yield Calculator

  1. Click Load Example to see a sample purification table, or enter your own data.
  2. For each purification step, enter the step name, total volume (mL), protein concentration (mg/mL), and activity (units/mL) if available.
  3. The first row must be Step 1 — the crude extract or cell lysate. All recovery and fold-purification values are calculated relative to this step.
  4. Click Calculate Protein Yield to generate the full purification table with total protein, total activity, specific activity, % yield, and purification fold.

Understanding the Purification Table

Total protein (mg) = concentration (mg/mL) × total volume (mL). Tracks how much total protein remains after each step.

Total activity (units) = activity (units/mL) × total volume (mL). Tracks how much of the target enzyme or protein activity is retained.

Specific activity (units/mg) = total activity ÷ total protein. This should increase with each purification step — if not, that step is not enriching the target protein.

Yield (%) = (total activity at step N ÷ total activity at step 1) × 100. Shows how much of the original activity was retained.

Purification fold = specific activity at step N ÷ specific activity at step 1. Indicates how many times purer the protein is relative to the crude extract.
