Track protein yield, recovery %, specific activity, and purification fold across every step of your purification protocol. Generate a complete purification table instantly.
Enter data for each purification step. The first row should be the crude lysate (Step 1). All subsequent rows are purification steps (e.g. ammonium sulfate precipitation, affinity chromatography, etc.).
| # | Step Name | Total Volume (mL) | Protein Conc. (mg/mL) | Activity (units/mL) |
|---|
Activity units/mL = enzyme activity per mL (e.g. nmol/min/mL, U/mL). Leave blank if you only want yield/recovery without specific activity.
| Step | Total Vol (mL) | Total Protein (mg) | Total Activity (units) | Specific Activity (units/mg) | Yield (%) | Purification Fold |
|---|
Total protein (mg) = concentration (mg/mL) × total volume (mL). Tracks how much total protein remains after each step.
Total activity (units) = activity (units/mL) × total volume (mL). Tracks how much of the target enzyme or protein activity is retained.
Specific activity (units/mg) = total activity ÷ total protein. This should increase with each purification step — if not, that step is not enriching the target protein.
Yield (%) = (total activity at step N ÷ total activity at step 1) × 100. Shows how much of the original activity was retained.
Purification fold = specific activity at step N ÷ specific activity at step 1. Indicates how many times purer the protein is relative to the crude extract.
